摘要:
饰纹姬蛙(Microhyla fissipes)在中国广泛分布于秦岭以南地区,在四川盆地是常见的两栖动物之一,研究其种群遗传多样性为当地两栖动物的保护与管理提供了重要的遗传基础资料和理论依据。本文用线粒体COI基因为分子标记,探讨了四川盆地饰纹姬蛙种群遗传多样性及其分化程度、种群历史动态。共采集饰纹姬蛙28种群354只,成功获得其COI基因618 bps片段序列。分析结果发现17种单倍型,总体单倍型多样性(Hd)为0.166、核苷酸多样性(Pi)为0.000 75,基因流(Nm)为1.60。各种群间固定系数(Fst)和遗传分化系数(Gst)分别为0.270、0.140。中性检验中,Tajima’s D = -2.380 ,P<0.001;Fu’s Fs = -22.169,P<0.02,均具有显著性差异;错配分布图呈泊松状的单峰。通过计算得出种群扩张时间约在更新世晚期0.235 MP(百万年)。结果表明,四川盆地饰纹姬蛙种群遗传多样性较低、种群间基因交流水平高,不同种群间遗传分化不明显,并且在历史上有过扩张的现象。(图5 表2 参47)
Abstract:
Microhyla fissipes is widely distributed in the south of Qinling Mountains in China, especially in Sichuan Basin, which was considered to be one of the common species. It is of significance for us to provide genetic data and theoretical basis of Microhyla fissipes for the process of conservation and management in other local amphibian. To study about the genetic diversity, population differentiation and history of Microhyla fissipes in Sichuan basin , the mitochondrial COI gene was used as a molecular marker . 618 bps fragments of COI gene were successfully sequenced in 354 individuals sampled from 28 populations in Sichuan Basin. A total of 17 haplotypes were identified. The average haplotype diversity (Hd) and nucleotide diversity (Pi) were 0.166 and 0.000 75, respectively. The gene flow ( Nm) among populations was 1.60. The coefficients (Fst) and genetic differentiation coefficient (Gst) of various groups were 0.270 and 0.140, respectively. The differences of the Neutral test (Tajima’s D values = -2.380, P<0.001 and Fu’s Fs values = -22.169, P<0.02) was significant. And the mismatch distribution maps showed a single peak of Poisson,which indicated that M. fissipes has undergone population expansion recently . T he population expansion time was calculated to be about 0.235 MP (millions of years) in the late Pleistocene. The results indicated that the population genetic diversity was relatively low, while the level of gene exchange between populations was contrarily to be high. In this case, no obvious differentiation among diverse populations in Sichuan Basin was observed . Actually,the expansion in Microhyla fissipes had been reported in the past.
