摘要:
为促进干秸秆的跨季节贮存及尾菜资源化利用,利用青贮原理将干玉米秸秆与白菜废弃物进行混合贮存发酵,探讨了不同添加量乙酸对二者混贮发酵品质的影响,并通过Miseq高通量测序技术解析发酵过程中的微生物多样性。试验设置ME组(无乙酸添加)为对照组,AA组(添加量0.3%)和AB组(添加量0.6%)为2个乙酸处理组,18±1 ℃恒温密闭混贮60 d,间隔30 d分析其化学组分、发酵品质及微生物多样性。结果表明,AA组贮存60 d时的干物质(DM)、可溶性碳水化合物(WSC)和粗蛋白(CP)含量均显著高于ME组,酸性洗涤纤维(ADF)和酸性洗涤木质素(ADL)含量显著下降,半纤维素(HC)、纤维素(CL)和综纤维素(HOC)含量显著增加。AB组在30d和60d时pH均显著低于ME组,乳酸含量在30 d时显著增加,60 d时显著降低。AA组和AB组中乳酸/乙酸、乳酸/总有机酸及氨氮/总氮值均低于ME组。混贮发酵期间ME组、AA组和AB组的门水平细菌主要为Proteobacteria和Firmicutes,属水平细菌包括Lactobacillus、Paralactobacillus、Enterobacter、Pediococcus、Flavobacterium、Chryseobacterium、Pedobacter、Sphingomonadaceae、Erwinia等,其中Lactobacillus、Paralactobacillus和Enterobacter为优势菌。与ME组相比,AA组和AB组中腐败菌Enterobacter丰度呈下降趋势;2个乙酸处理组的总乳酸菌丰度高于ME组,但乳酸菌多样性较ME组有所减少,主要包括Lactobacillus、Paralactobacillus和Pediococcus等乳酸菌属。总之,干玉米秸秆与白菜废弃物能以适宜比例混贮60d不变质,尽管添加乙酸后的乳酸发酵强度有所抑制,但低剂量乙酸更有助于保存DM、WSC、CP、HOC等有机组分,建议乙酸添加量为0.3%。(图3,表5,参42)
Abstract:
In order to realize the trans-seasonal storage of dry maize straw (DMS) and utilization of vegetable wastes, the mixed airtight wet-storage of DMS and cabbage waste (CW) were implemented at 18±1℃ for 60 days based on the silage theory. The dynamic effects of different doses of acetic acid on the storage and fermentation quality were investigated, and the dynamic rule of microbial community diversity during storage were also investigated by Illumina MiSeq high-throughput sequencing. Group ME (without the addition of acetic acid), group AA ( the addition of acetic acid with the dosage of 0.3%) and group AB (the addition of acetic acid with the dosage of 0.6%) were set, the changes of the chemical composition, fermentation quality and microbial community diversity were analyzed for the interval of for 30days. The experimental result showed that the contents of dry matter (DM), water soluble carbohydrates (WSC) and crude protein (CP) were significantly (p<0.05) higher than those in group ME at 60 d, the contents of acid detergent fiber (ADF) and acid detergent lignin (ADL) were significantly (p<0.05) decreased compared with group ME, the contents of hemicellulose (HC), cellulose (CL) and holocellulose (HOC) in group AA were significantly increased compared with group ME (p<0.05). The pH value in group AB was significantly (p<0.05) lower than that of group ME at 30 d and 60 d, the contents of lactic acid were increased significantly (p<0.05) at 30 d, and then decreased significantly (p<0.05) at 60 d compared with group ME. As a result, the ratio of lactic acid and acetic acid (LA/AA), the ratio of lactic acid and total organic acid (LA/TOA) and the ratio of ammonia nitrogen and total nitrogen (AN/TN) in group AA and AB were lower than that of group ME. The dominant bacterium communities in three groups were Proteobacteria and Firmicutes at phylum. The bacterium communities at genus including Lactobacillus, Paralactobacillus, Enterobacter, Pediococcus, Flavobacterium, Chryseobacterium, Pedobacter, Sphingomonadaceae and Erwinia, Furthermore, Lactobacillus, Paralactobacillus and Enterobacter were dominant bacterium communities. The abundance of Enterobacter was demonstrated decreased tendency during mixed storage period. The abundance of total lactic acid bacteria in group AA and AB were higher than that of group ME, but the lactic acid bacteria diversity in group AA and AB were lower than that of group ME, and the main lactic acid bacteria were Lactobacillus, Paralactobacillus and Pediococcus. In conclusion, DMS and CW with optimum proportion can be mixed storage by ensiling fermentation for 60 d with good quality, the storage quality were further improved by the addition of acetic acid, the addition of acetic acid with the dose of 0.3% can be proposed and conducive to the preservation of the organic components, such as DM, WSC, CP and HOC.
