摘要:
为得到高效的苯甲酰脲类杀虫剂降解菌,为其污染修复的开展提供资源和理论基础,从农药厂活性污泥中分离筛选到一株可降解灭幼脲、除虫脲、氟铃脲的菌株,命名为M6。经生理生化特征和16S rRNA序列分析,将其鉴定为无色杆菌属(Achromobacter sp.)。菌株M6可在48 h内降解90%以上初始浓度为100 mg?L-1的灭幼脲、除虫脲、氟铃脲;且可在不添加其它碳源的情况下,以这3种杀虫剂为唯一碳源生长。选取菌株降解效果最好的灭幼脲为底物,研究其降解特性。菌株M6降解灭幼脲时,对温度、pH值等培养条件适应范围较宽,降解灭幼脲的最适温度为30 ℃,最适pH为7.0;可耐受400 mg?L-1的灭幼脲。通过对乙酰氨基酚变色和简并引物克隆的方法,初步确定菌株M6通过水解酰胺键降解灭幼脲、除虫脲、氟铃脲。(图8 参30)
Abstract:
Objectives: To provide new resource and valuable information of benzoylurea insecticides degradation, a chlorbenzuron, diflubenzuron, and hexaflumuron-degrading bacterium strain M6, was isolated from the activated sludge of an insecticide factory. This paper aimed to study the biodegradation characteristics and pathway of M6.Methods: The strain was identified according to analysis of 16S rRNA gene sequence, morphological and physiological characteristics. The degradation characteristics of strain M6 to 3 benzoylurea insecticides under various conditions, and cloning of arylamidase gene of strain M6 were also investigated. Results: Strain M6 was identified as Achromobacter sp., and could degrade more than 90% of 100 mg昄-1 cohlorbenzuron, diflubenzuron, and hexaflumuron within 48 hours, which could be used as sole carbon source. M6 displayed better effect for chlorbenzuron degradation at temperatures ranging from 25-40 ℃ and at pH ranging from 5.0 to 9.0. The optimal temperature, initial pH of medium for chlorbenzuron degradation by M6 were 30 ?C and 7.0, respectively; the maximum chlorbenzuron tolerated concentration for M6 was as high as 400 mg昄-1. M4-acetaminophenol could be hydrolyzed into purple-red product by M6. Moreover, M6 contained approximately 1.4 kb DNA fragment, which can be expressed into an amidase to degrade amide pesticides.Conclusions: The relatively high degradation rate was achieved, and 3 benzoylurea insecticides could be degraded owing to their amide bonds-hydrolyzing by M6.
